Zusammenfassung
Seit der Erstbeschreibung der Plaques und der neurofibrillären Degeneration durch A. Alzheimer als morphologisches Korrelate der Alzheimerschen Demenz (AD) zu Anfang des Jahrhunderts versuchten verschiedene retrospektive und prospektive Studien den Zusammenhang zwischen Pathomorphologie und Demenz zu klären [3, 6–8]. Untersuchungen, die den Nachweis des β-Amyloid-Proteins als Hauptbestandteil der Plaques zum Gegenstand hatten, konnten keine gute Korrelation zwischen der Beladung des Parenchyms mit Amyloid und der Demenz nachweisen. Amyloidablagerungen fanden sich in zum Teil massiven Ausmaß auch bei nichtdementen älteren Personen. Erste Hinweise auf ein entzündliches Geschehen in den Plaques von dementen Personen ergaben sich aus dem Nachweis des Akut-Phase-Proteins a1-Antichymotrypsin und aktivierter Mikrogliazellen [1, 11]. Wir konnten erstmalig das Cytokin Interleukin-6 (IL-6) immunhistochemisch in Plaques in Gehirnen von Alzheimer Patienten nachweisen [4, 14]. Dieser Nachweis konnte mittlerweile durch biochemische Methoden bestätigt werden [16].
