Abstract

Acoustic neuroma is a benign peripheral nerve sheath tumor of the eighth cranial nerve arising from Schwann cells. Although commonly used, the term “acoustic neuroma” is inaccurate because this neoplasm does not derive from the acoustic branch of the VIII cranial nerve (but rather from the vestibular branch) and is not a neuroma.

    NF2: this gene encodes neurofibromin 2, a protein that is similar to some members of the ERM (ezrin, radixin, moesin) family of proteins that are thought to link cytoskeletal components with proteins in the cell membrane.

    NF1: this gene encodes neurofibromin 1, a negative regulator of the RAS signal transduction pathway. Neurofibromin acts by accelerating the conversion of active GTP-bound RAS to its inactive GDP-bound form. The RAS signaling can be activated by receptor tyrosine kinases following the binding of growth factors, which results in increased AKT and/or MEK activity. In addition, RAS controls the generation of cyclic AMP (cAMP) through protein kinase C (PKC) following the activation of a G protein-coupled receptor (GPCR).
