Galectin-3 expression in various thyroid neoplasms and its possible role in metastasis formation*

Abstract

Galectin-3 is a member of the β-galactoside-binding protein family that plays an important role in cell-cell adhesion and in cell-matrix interaction. We have examined the expression of galectin-3 in normal, adenomatous, and malignant thyroid tissues and also in metastasic lesions. Galectin-3 was rarely expressed in normal thyroid tissue but was abundant in the cytoplasm of the neoplastic lesions. Among neoplastic lesions, galectin-3 was expressed to a greater extent in follicular carcinomas than in follicular adenomas and was present in greater amounts in papillary carcinomas than in follicular adenomas or carcinomas. Primary lesions of papillary carcinoma with metastasis contained significantly higher concentrations of galectin-3 than tumors of this type without metastases. However, the expression of galectin-3 was significantly decreased in metastasic lesions in the lymph nodes compared with their primary lesions. From these results, we assumed that galectin-3 works in different ways at different stages of thyroid neoplasm proliferation. Among primary tumors, galectin-3 expression is significantly different in 3 histological types. However, the continuity of progression among these tumors is not yet proven. In later stages, decreased expression of galectin-3 may aid the release of cancer cells from the primary lesions for invasion and metastasis.

      This association can be related with the cell migration properties of galectin-3 in the early stages of colorectal cancer development. In addition, it has been suggested that the decreased galectin-3 may loosen the connection of cancer cells with the extracellular matrix to facilitate the invasive and metastatic properties of cancers [22]. A similar effect can be attributed to the expression and correlations between galectin-4, 7, 8, and 10 mRNAs, which showed slightly increased levels of their mRNA expression in colorectal carcinomas without any lymph node or distant metastasis.

      Turkoz and colleagues debated these findings, where the authors demonstrated a lower expression of galectin-3 in lymphatic metastases and their primary carcinomas (Türköz et al., 2008). It has been suggested that the decreased galectin-3 may loosen the connection of cancer cells with the extracellular matrix to facilitate the invasive properties of cancers (Kawachi et al., 2000). The results presented here do not support this latter theory, rather suggesting that an increased level of galectin-3 expression may be facilitative for cancer metastasis where the protein may act to modulate cell growth and be involved in the regulation of apoptosis (Dumic et al., 2006; Newlaczyl and Yu, 2011).
